Since late 2011, porcine infections with highly virulent and antigenic variant of pseudorabies virus (PRV) cause great economic loss in the swine industry in China, and its emergence leads to variable protection efficacy of the commercially available PRV vaccine.
In the present study, the potential cross-protective efficacy of two live virus vaccines, includ- ing a commercial vaccine, and an attenuated low pathogenic PRV variant (rPRVTJ-delTK/gE/gI) against a PRV variant Tianjing (TJ) was evaluated in piglets. Vaccination of piglets with the live vaccine Bartha-K61 could not reduce the clinical signs, and was partially efficacious in the reduc- tion of viral loads upon PRV variant TJ challenge, indicating that this live vaccine provided limited cross-protection efficacy against the PRV variant infection. Additionally, rPRVTJ-delTK/gE/gI appeared to exert some beneficial efficiency in shortening the period of clinical fever and improv- ing the growth performance of the challenged pigs.
Our findings give a valuable guidance for the choice and use of PRV vaccines to control PRV variant infection in the field.
Vaccination is a common routine for prevention and control of human and animal diseases by inducing antibody responses and cell-mediated immunity in the body. Through vaccinations, smallpox and some other diseases have been eradicated in the past few years. The use of a patho- gen itself or a subunit domain of a protein antigen as immunogens lays the basis for traditional vaccine development. But there are more and more newly emerged pathogens which have expe- rienced antigenic drift or shift under antibody selective pressures, rendering vaccine-induced im- munity ineffective. In addition, vaccine development has been hampered due to problems includ- ing difficulties in isolation and culture of certain pathogens and the antibody-dependent enhancement of viral infection (ADE). How to induce strong antibody responses, especially neu- tralizing antibody responses, and robust cell-mediated immune responses is tricky. Here we re- view the progress in vaccine development from traditional vaccine design to reverse vaccinology and structural vaccinology and present with some helpful perspectives on developing novel vac- cines.
Based on the rolling bearing vibration measurement principle in ISO standard, a nonlinear dynamic model of ball bearing is built and motion equations of the inner ring, outer ring, and rolling elements are derived by using Lagrange’s equation. The ball bearing model includes the influence of waviness, rotational speed, external load, arbor supporting stiffness and arbor eccentricity. Ball bearing high-speed vibration tests are performed and used to verify the theoretical results. Simulated results showed that specific waviness orders produced the principal frequencies that were proportional to rotational speed. Rotational speed mainly affected the value of the natural frequency of the bearing system, and RMS (Root Mean Square) of the full band had a great fluctuation with the increase of rotational speed. In the experiment, spectrum and RMS of 2fs-30 kHz (fs: the rotational frequency of inner ring/arbor) under high speed could include not only the influence of rotational speed but also principal frequencies produced by waviness, which could cover the part of requirements of the standard bearing vibration measurement.
